一、揭示了小麦‘绿色革命基因’的分子基础、提出了赤霉素“去抑制促生长”理论

赤霉素是一种重要的植物激素,对植物生长发育起关键的调节作用。但直至上世纪90年代,人们对赤霉素信号传导的机制几乎一无所知。自1990年以来彭金荣教授在该领域开展了系统的研究工作:

1 小麦半矮秆基因Rht也被称为‘小麦绿色革命基因’,这是因为自育种学家Norman BorlaugRht引进小麦栽培种后全世界的小麦产量大幅提高,从而解决了发展中国家(如墨西哥、巴西等)的粮食问题。目前世界上超过90%的栽培小麦品种带有Rht基因。由于Norman Borlaug在粮食‘绿色革命’中的重大贡献,他在1970年获得了诺贝尔和平奖。上世纪90年代彭金荣教授经过多年的努力首先克隆了模式植物拟南芥的半矮秆基因gai,然后通过比较基因组学方法克隆了小麦中gai的同源基因Rht基因,从而在分子水平上揭示了‘小麦绿色革命基因’的本质。同时他还克隆了玉米中gai的半矮秆同源基因D8

2 进一步的研究显示gai/Rht/D8编码植物激素赤霉素信号传导的负调控因子,现被统称为DELLA蛋白。赤霉素通过诱导DELLA蛋白的降解来促进种子萌发、茎秆生长、花期诱导以及雄配子发育。根据自己的工作彭金荣教授及其同事们提出了广为接受的赤霉素“去抑制促生长”的作用机制,这个理论主导了近十年内研究人员对植物激素赤霉素(GA)信号传导的研究,为人们认识赤霉素作用的分子机制作出了重要贡献。值得一提的是该理论已成为植物生理学教科书(Plant Physiology, Fifth Edition, Lincoln Taiz & Eduardo Zeiger, 2010)中赤霉素信号传导章节的重要组成。

在赤霉素信号传导领域,彭金荣作为通讯作者或共同通讯作者或第一作者发表了18篇与赤霉素相关的有重要影响的系列论文(发表在包括Nature x1, Science x1, Genes & Development x2, Plant Cell x1, Plos Genetics x1, Development x1, Plant J x1, Plant Physiology x3等期刊上),这些论文的总引用次数已达2600余次(查自SCI核心期刊集2015.01.12)。

 

二、发现了核仁中以Def-Calpain3为轴心的新的蛋白降解途径

肝脏是体内重要器官。尽管人们对肝脏的生理功能已有深入了解,但至今对于肝脏早期发育的分子调控机制却知之有限。自2000年起彭金荣实验室以斑马鱼为模式动物系统地开展了肝脏发育的研究:

1 Def是彭金荣实验室首先研究并命名的一种新型核仁蛋白。在斑马鱼中def基因失活导致肝脏变小。通过研究def 突变体他的实验室发现了肿瘤抑制因子p53的异构体Δ113p53,并发现def突变体中p53的激活直接诱导Δ113p53转录表达,而Δ113p53蛋白的功能是促进p53抑制细胞周期的功能、拮抗p53促细胞凋亡功能。这些研究结果揭示了一个新的调控肝脏发育的途径,并开辟了p53研究的新领域。

2 回国后的主要学术贡献:

进一步的研究发现Def与半胱氨酸蛋白酶Calpain3形成复合体介导p53的降解。这些研究结果证明在核仁中存在一个由Def-Calpain3 为轴心的介导蛋白降解的新途径来调控细胞增殖与死亡,并由此调控肝脏的发育。该研究成果将极大地拓宽我们对核仁生物学功能的理解,同时将为细胞生物学和发育生物学开辟新的研究方向。同时,彭金荣实验室与合作实验室进一步研究了p53的异构体Δ113p53/Δ133p53的生物学功能,发现Δ113p53/Δ133p53直接参与激活DNA双链断裂修复关键基因的表达,从而解决了长期困扰本领域有关p53如何参与DNA双链断裂修复的疑惑。

在斑马鱼发育研究领域彭金荣作为通讯或共同通讯作者发表了24篇系列论文(发表在包括Genes & Development x2, Cell Research x2, Genome Research x1, Blood x1, Development x1, Nucleic Acid Research x 1, Developmental Biology x2, JBC x1等期刊)。这些论文的总引用次数达420余次(查自SCI核心期刊集2015.01.12)。
